To disconnect male luer lock from a female one, you should first place the projecting part of the cap over point 16a (this secures luer withing minimally increasing diameter pulled past orifice) and rotate it into this position. When you bring them together, give the male lock a twist or two to assure snugness. Once that is complete, place the two pieces together and twist a few more times on the male luer lock until it feels secure. But be warned that you should not screw it in too hard or else risk breaking a piece. To disconnect them later, loosen the male luer lock by turning it several times in the opposite direction.
The problem that often occurs with luer lock connections are they can become loose over time and you will end up leaking. If you experience this, tighten the connection a little more but be careful not to overdo it. If that luer lock breaks it is really important you fix that as soon as possible because bad things can happen. Also, double-check that you are using the correct luer lock device with the equipment in which you work.
